Yeast strain PJ69-4A containing a plasmid expressing a GAL4BD:: RAD7 fusion gene (TRP1 selection) was transformed with a mixed library, made in plasmids pGAD-C1, pGAD-C2, and pGAD-C3 (LEU2 selection (Philip James, personal communication) using Protocol 2. These numbers are from a single transformation reaction. There has been NO scale up! The transformants were plated onto SC omission medium minus Trp, Leu medium.
